GyrA and ParC Gene Analysis of the Quinolone Antimicrobial Susceptibility-decreased Serratia marcescens Strains Isolated from Contact Lens Case

Abstract
Purpose: For Serratia marcescens strains appeared to quinolone antibiotic susceptibility-decrease used in the treatment of bacterial keratitis by analyzing the main target gyrA and parC genes involved in resistance mechanisms and to investigate the cause of the susceptibility decrease.

Methods: By extracting genomic DNA targets of quinolone antimicrobial susceptibility-decreased Serratia marcescens strains after amplification by PCR and analyzed by DNA sequencing of the gyrA and parC gene QRDR.

Results: In the parC gene analysis, SM5 strain showed double amino acid mutations (Tyr51¡æHis, Thr59¡æAsp), SM6 strain showed (Thr59¡æAsp), SM13 strain showed single amino acid mutation (Tyr51¡æHis). And 3 strains showed many silent mutations in gyrA and parC gene.

Conclusions: It could be confirmed that the antimicrobial susceptibility caused by mutations in target gene is lowered, especially parC gene was confirmed the major target causes sensitivity degradation of quinolone antibacterial agents. We will continue the use of quinolone antimicrobial in the treatment of bacterial keratitis, many resistant strains are predicted would occur. Therefore, when using the antimicrobial is always careful to be necessary in resistant.
